Sensitivitas Real-Time Polymerase Chain Reaction dengan Primer Tanabe dalam Mendeteksi Gelatin Babi pada Confectionery

Gelatin is commonly used as a gelling and thickening agent in confectionery products considered critical material terms of its halal status. Real-time Polymerase Chain Reaction (real-time PCR) frequently employed an analytical tool to detect traces pork food items. Although the real-time PCR method using Tanabe primer Internal Positive Control (IPC) has been validated effectively, sensitivity or Limit Detection (LOD) for yet be determined. This study aims determine with primers IPC towards (gummy candy, marshmallow, lozenges). Confirmation this on commercial marshmallow known contain porcine gelatin was also carried out. In study, LOD (% w/w) bovine initially The findings revealed that 0.01% cycle threshold (Ct) value 39.20±1.72. next step involved determining various such lozenges, gummy marshmallows. lozenges candy found 0.1% Ct values 40.93±0.15 38.72±0.18, respectively. Marshmallows exhibited 41.14±2.96. Finally, applied containing gelatin, effectively detected high sensitivity.
